Nothing else is active. The other ingredients are the coating and binders plus potato starch. The extract comes from pork, so the tablet is not vegetarian, and the brand’s own FAQ says it may not suit anyone who avoids pork. What DAO is, and how a tablet is meant to act
Diamine oxidase is an enzyme made by the lining of the small intestine that breaks down histamine from food. The idea is that extra enzyme taken before a meal can break down some of the histamine in aged cheese, wine, fish or leftovers before it reaches the blood, and that the enteric coating keeps stomach acid from destroying it first. The brand says the tablet does not activate until the small intestine and shows no release data. The trials below measured symptom scores, not histamine in the blood after a real meal.
The trials of DAO tablets
We could find no trial of HistaHarmony: PubMed returned nothing for the product name or ProDAOX, and ClinicalTrials.gov listed no study. The page’s “Clinically-Studied” badge links to no paper. The evidence is for DAO supplements as a class:

Only the first two trials concern food-histamine symptoms, and one is open label. The migraine, hives, fibromyalgia and insomnia trials tested other conditions, with mixed results. We found no trial of a DAO tablet against reactions to ordinary meals in people whose condition a placebo-controlled challenge had confirmed.
A diagnosis that is hard to confirm
Histamine intolerance has no accepted diagnostic test. In a 2023 placebo-controlled challenge study from Munich, 62.7% of 59 patients with suspected histamine intolerance reacted to placebo, and the condition was excluded in 50 of them (84.7%). Blood DAO activity varied widely in those without the condition, which the authors said limits its value as a marker.
What 30,000 HDU means
HDU is an activity unit the brand reports, not a standard dose. Other Amazon listings print 20,000 HDU per serving on one DAO product and 1,000,000 HDU per tablet on another, so a larger number on one box is not a like-for-like comparison with another label.
How to take HistaHarmony, and how the dose compares
The label says: take one tablet just before histamine-rich foods or on an empty stomach, as a healthcare practitioner recommends, swallowed whole, 20 minutes before meals. The brand page adds one tablet away from meals for general support, such as at bedtime, and says changes can take several days to a few weeks. No daily maximum is printed.
That is one third of the dose in the fibromyalgia and insomnia trials, which gave three 4.2 mg tablets (12.6 mg) a day; the open-label pilot allowed up to three capsules. A tablet before each of three meals would reach that amount, but the label does not say to.

On Amazon the listing shows 4.4 stars from 306 ratings, with 76% five-star and 8% one-star. Amazon’s review summary counts 62 positive and 13 negative mentions of effectiveness, and 25 positive and 17 negative on side effects. The recurring praise is tolerating foods that used to cause reactions, relief of hives and flushing, and tiny tablets. The recurring complaints are reactions after taking it, including hives, brain fog, nausea and bloating, and doubt about whether it works.
The brand’s Trustpilot profile (4.5 from 23 reviews) covers the whole company, as does its review vendor’s store score. The brand page shows no star count of its own, only a survey (97% tolerated it well) with no respondent count and no comparison group.

Both are DAO enzyme supplements, and neither has a trial of its own formula. HistaHarmony is a single ingredient, 4.2 mg of porcine kidney extract at 30,000 HDU in a delayed-release tablet.
